Incidence of Mouse Bladder Tumors following Implantation of Paraffin Pellets Containing Certain Tryptophan Metabolites.

چکیده
Pellets of paraffin wax implanted into the lumen of the bladder of 38 mice induced one carcinoma. An equally low incidence of carcinomas was induced with paraffin wax pellets containing 15-20 per cent suspensions of 8-hydroxyquinoline or quinaldic acid, 8-hydroxyquinaldic acid, xanthurenic acid, the 8-methyl ether of xanthurenic acid, 4,8-quinolinediol, L-kynurenine sulfate, 3-hydroxy-L-kynurenine, and 3-hydroxyanthranilic acid, all metabolites of tryptophan. The inability to produce pellets of uniform composition containing these test substances was demonstrated. Paper Chromatographie methods revealed that most of these compounds remained in the paraffin vehicle in vivofor as long as 342 days. It was concluded that paraffin is not a satisfactory vehicle in which to test these urinary tryptophan metabolites for bladder carcinogenicity.

Mouse Bladder Carcinogenicity of Certain Tryptophan Metabolites and Other Aromatic Nitrogen Compounds Suspended in Cholesterol.
The carcinogenicity of seven metabolites of the essential amino acid tryptophan and nine other nitrogen-containing aromatic compounds was studied by the individ ual suspension of these chemicals in cholesterol pellets and the surgical implantation of the pellets into the bladders of mice. Pellets consisting of the 8-methyl ether of xanthurenic acid, xanthurenic acid, 8-hydroxyquinaldic acid, 3-...

Several compounds—arachic acid, hexaethylbenzene, hexamethylbenzene, 1-octadecanol, palmitic acid, and stearamide— were individually compressed as pellets and tested, by im plantation into the mouse bladder, as possible vehicles for carcinogenicity experiments. All of these compounds were found to b3 associated with a 3-17% incidence of bladder carcinomas. Pellets of 23-methylcholanthrene (...
متن کاملEffect of the Urine and Calculus Formation on the Incidence of Bladder Tumors in Rats Implanted with Paraffin Wax Pellets1
A double pouch of bladder was formed by divison of the bladder of inbred Fischer 344 rats and implantation of paraffin pellets into each pouch. The upper pouch could be in contact with or isolated from the urinary stream, while the lower pouch was always in contact with the urine. No tumors formed in 99 isolated upper pouches, while 49 formed in 119 communicating upper pouches in contact with t...

To test the hypothesis that o-hydroxy amines are proximate agents in chemical carcinogenesis paraffin pellets containing the hydrochlorides of 2-amino-l-fluorenol, 2-amino-3-fluorenol, 2-amino-5-fluorenol, 2-amino-7-fluorenol, 2-fluorenamine, and 1amino-2-naphthol were implanted into the urinary bladders of Swiss female mice. Paraffin pellets alone were implanted as controls. Histological exami...
متن کاملTryptophan metabolites in the urine of Turkish cows with urinary bladder cancer.
Industrial urinary bladder cancer in dye workers has been shown to be caused by exposure to certain aromatic amines (cf. 21 for review). Animal experiments have indicated that it is likely that the carcinogens are o-aminophenol metabolites (4, 5) and that these metabolites probably are carried to the urinary bladder by the urine (26, 41). Unidentified diazotizable aromatic amines in the urine o...
